Take home problem to be worked with Ionex.

You can complete the following take home problem and turn in your results by e-mail or on a paper copy when you turn in your report.
To begin this project, you will need to download and install Ionex.zip. This file contains the program, help files, some protein files and some sample separation files (these end in .iex). It works best if all the files are installed in the directory "C:\ionex". You will need to use Winzip to extract these files. In addition to the ionex program, the zip file also contains the files for the four proteins you will be studying:

Glycogen phosphorylase (filename = 1pyg.pdb)

Hemoglobin (filename = 1hga.pdb)

cathepsin (filename = 1the.pdb)

Tumor suppressor P53 (filename = 1tup.pdb)

To receive credit, you will need to do the following:

Select conditions that will separate the four proteins into four separate peaks on the chromatogram. Conditions include the buffer selected, salt concentration in solvent A and solvent B and stationary phase. You can either write these on a piece of paper, or save your designed experiment using the "Save as" button on the Design Experiment window.
Identify the isoelectric point for each of the four proteins using the molecular view window. You need to adjust the pH bar to get the charge as close to 0.0 as possible. Report the pH value that you have found. The scale on the slider bar has 14 tick marks for pH values ranging from 1 to 14
