The detector is used to identify protein bands in the eluent from the column. Detectors are selected based on their ability to identify proteins in solution. The UV-Visible detector is most commonly used based on its ability to see proteins at wavelengths where they absorb strongly, typically, 214 nm, 254 nm or 280 nm. Fluorescence detectors are also used commonly to detect proteins in column eluates. The model system in this application has a UV-Visible detector set at 280 nm.
